A2.2.2: DIGESTION OF PROTEINS AND POLYSACCHARIDES VIA BIOCATALYSIS AND CHARACTERIZATION OF BIOACTIVE PEPTIDES FROM BEACH-CAST MACROALGAE BIOMASS

 

DESCRIPTION:

  1. Collection of algae from both the foreshore and the intertidal zone; identification and separation of species; extraction procedures (via trituration, filtration, centrifugation); quantification of total proteins and polysaccharides; storage at -80ºC
  2. Controlled enzymatic digestion; separation of peptides by molecular exclusion chromatography (<3000 Da) and/or ultracentrifugation
  3. Separation and identification of polysaccharides by chromatographic techniques
  4. Selection of hydrolysates and polysaccharides with antimicrobial, antioxidant, immunomodulating, angiotensin II inhibiting or anticancer activities through in vitro assays and tests on model organisms (Caenorhabditis elegans and Drosophila melanogaster)
  5. HPLC fractionation of the bioactive hydrolysates until the purification of the active peptides; MS sequencing

 

EXPECTED RESULTS

  1. Identification of peptides and/or polysaccharides with activities of interest for their application in pharmacology or biotechnology (antioxidant, immunomodulatory, antitumor, antimicrobial, insecticide, molluscicide, nematicide, anti-aging activity)
  2. Characterization of these molecules and of their mode of action by MS and chromatographic techniques
  3. Optimization of the extraction, separation and purification processes to facilitate the industrial use of the identified molecules of interest
  4. Transfer of results to companies
